Does an e-book qualify as a trademark specimen for a book?

No, an e-book wouldn’t qualify as a specimen if your trademark application specifies actual physical books in the goods and services. Furthermore, e-books and physical books are classified in different Nice Classification classes: e-books in Class 9, and physical printed books in Class 16. A specimen submitted for one class does not satisfy the requirements for the other.

If your trademark application covers both types of books and both classes, you must submit a separate specimen for each class showing the trademark in active use in connection with the goods in that specific class.
